چکیده

We studied the number and affinity of catecholamine receptors in SD rat kidney by radioreceptor technique. The following conclusions were obtained: 1) By using 3H-prazosin, the numbers of alpha 1-receptor (Bmax) in rat renal cortex were greater than those in rat kidney medulla. As for affinity (Kd), the significance was not recognized between the two. Bmax of the rat renal cortex to 3H-prazosin binding was 96.1 fmol/mg protein, and Kd was 0.17 nM, and for the rat renal medullar these values were 44.5 fmol/mg protein and 0.13 nM, respectively. 2) By measurement of D1-receptor using 3H-SKF38393 in the rat renal cortex in the Scatchard plot analysis, positive cooperativity was observed under the low concentration of hot ligand which was less than 1 nM. But at the concentration of hot ligand over 1 nM, the plots showed a straight line. Bmax of the rat renal cortex to 3H-SKF38393 was 2.5 pmol/mg protein and Kd was 5.3 nM. 3) Based on displacement by dopamine for 3H-prazosin binding to rat renal cortex, it was surmised that high concentration of dopamine had an affinity to alpha 1-adrenoceptors. 4) There was no change in the Kd and Bmax of alpha 1-receptor in the rat renal cortex after incubation of samples with low concentration of dopamine. However, in the case of high concentration of dopamine, a remarkable decrease of the affinity (Kd) of alpha 1-adrenoceptor was observed.

Dopamine inhibits vasopressin action in the rat inner medullary collecting duct via alpha(2)-adrenoceptors.

We compared the effects of dopamine and norepinephrine on vasopressin (AVP)-stimulated increases in osmotic water permeability (Pf) and cAMP accumulation in the rat inner medullary collecting duct (IMCD). Both dopamine and norepinephrine inhibited AVP-induced Pf and cAMP accumulation in a concentration-dependent manner; however, norepinephrine was approximately 100-fold more potent than dopamin...

Presence of prejunctional D2-dopaminoceptors and α2-adrenoceptors on the cholinergic nerve of the common bile duct of guinea pig

On most adrenergic and cholinergic nerve terminals, prejunctional α-adrenoceptors belonging to the α2-subtype have been identified. Activation of these receptors will decrease the release of norepinephrine. It has been reported that several isolated tissue preparations contain prejunctional dopamine receptors, the stimulation of which inhibits neurotransmission. It has remained uncertain whethe...

EFFECTS OF CATECHOLAMINES ON DOPAMINE AND SEROTONIN SYNTHESIS IN RAT BRAIN STRIATAL SYNAPTOSOMES: THE ROLE OF PRESYNAPTIC RECEPTORS AND THE SYNAPTOSOMAL REUPTAKE MECHANISM.

The regulation of dopamine and serotonin synthesis in rat brain striatal synaptosomes has been studied using HPLC methods. Noradrenaline was shown to markedly inhibit both the synthesis of dopamine and serotonin. The response of the synaptosomes to the concentrations of noradrenaline appeared to be biphasic, a very effective inhibition occurring at low concentrations (1-5 µm) and a relativ...

Effects of the stimulation of β2-adrenoceptors of the the basolateral amygdala on anxiety-like behaviors following electrical foot-shock stress in male rat

Introduction: The basolateral amygdala (BLA) is a key structure that integrated anxiety information. It is suggested that baso-lateral area of amygdala receives a dense norepinephrine innervation from the locus coeruleus nucleus. Furthermore, stress increased norepinephrine secretion in the BLA. In the present study, we examined the effects of the stimulation of β2-adrenoceptors in the BLA on a...

Preclinical pharmacology of fiduxosin, a novel alpha(1)-adrenoceptor antagonist with uroselective properties.

Benign prostatic hyperplasia (BPH), common in aging males, is often treated with alpha(1)-adrenoceptor antagonists. To minimize hypotensive and other side effects, compounds with selective antagonist activity at alpha(1A)- and alpha(1D)- (compared with alpha(1B)-) adrenoceptors were evaluated that would block lower urinary tract alpha(1)-adrenoceptors in preference to cardiovascular alpha(1B)-a...
